The VPl gene of enterovirus 71 (EV71) was synthesized, construct a recombinant plasmid pET15b/VP1 and expressed in E. coli BL21. The recombinant VP1 protein could specifically react with EV71-infected patient sera without the cross-reaction with serum antibodies of coxsackievirus A16 (CA16), A4, A5, B3 and B5 as well as echovirus 6. In acute and convalescent phases, IgM and IgG antibodies of 182 serum samples were detected by ELISA with recombinant VP1 protein as a coated antigen. Neospora caninum infection is an important disease affecting bovine and canine populations worldwide. The dense granule antigen 7 of N. caninum (NcGRA7) is considered to be an immunodominant antigen. In the present study, the gene encoding truncated NcGRA7 (NcGRA7t) lacking an N-terminal signal peptide was expressed in Escherichia coli, and its diagnostic potential in an enzyme-linked immunosorbent assay (ELISA) was evaluated. O objetivo deste estudo foi desenvolver uma vacina de DNA contra linfadenite caseosa, testá-la em camundongos BALB/c e avaliar a produção de anticorpos específicos e os níveis de proteção conferidos aos animais testados. The frequency of canine heartworm disease was determined in 310 dogs (158 males and 152 females) from São Paulo city (154) and several towns along the São Paulo state coast (156). The survey was carried out through the Knott's technique (direct presence of microfilariae in the blood) and ELISA for soluble circulating antigen detection. Twenty five (8.0%) samples were positive to D. immitis using both methods being 6 dogs (3.9%) from the city and 19 (12.2%) from the coastal area. Two methods were used to estimate the reproductive output of female Pacific oysters reared in Normandy: histology with image analysis and ELISA (Enzyme-Linked lmmunoSorbent Assay) which allowed the quantification of egg protein. Six bovines were infested with 60 first instar larvae of Dermatobia hominis. The animals were bleed weekly, and their antibodies levels to D. hominis L1, L2 and L3 instars measured during the time, following the infestation course. Durante las temporadas de 2001-2002 y 2002-2003 se realizó una prospección de virus en pimientos (Capsicum annuum L.) en diversas localidades de la Región de Coquimbo (29,5-32° lat. Sur). La determinación de virus se realizó mediante la prueba DAS- ELISA. Trypanosoma congolense is a major livestock pathogen in Africa, causing large economic losses with serious effects on animal health. Reliable serodiagnostic tests are therefore urgently needed to control T. congolense infection. In this study, we have identified one T. congolense protein as a new candidate serodiagnostic antigen. Babesia microti is a rodent tick-borne blood parasite and the major causative agent of emerging human babesiosis. Here, we identified a candidate of common antigenic protein BmP41 of B. microti by serological screening of cDNA library of human-pathogenic Gray strain with antisera against rodent Munich strain. Immunofluorescent antibody test using mouse anti-recombinant BmP41 (rBmP41) serum revealed that native BmP41 was expressed in each of the developmental stages of B. microti merozoites. ELISA was used to evaluate, follow, and compare the humoral immune response of Swiss mice during hyperimmunization with natural and Cobalt 60-irradiated (60Co) Crotalus durissus terrificus venom. Potency and neutralization were evaluated by in vitro challenges. After hyperimmunization, immunity was observed by "in vivo" challenge and the side effects were assessed. Paratuberculosis (Johne's disease) is a chronic enteritis in cattle caused by infection with Mycobacterium avium subsp. paratuberculosis (Map). In Brazil, few reports describe the isolation of the organism. The gold standard test for Map is its isolation from tissues or feces. Colheu-se sangue de sete suínos infectados com ovos de Taenia solium, semanalmente, durante 140 dias, para realizar ELISA no soro, utilizando antígeno de escólex (Es-Tso) de C. cellulosae. Em todos os animais, após o 21º dia pós-infecção, houve incremento significativo de anticorpos IgG, que assim se mantiveram até o final do experimento. A sensibilidade do ELISA variou entre 87,5 e 100%. À necropsia, foram identificados 238 cisticercos. Seis suínos apresentaram pelo menos um cisto no coração, língua ou masseter. Não se observou correlação entre concentração de anticorpos e número de cisticercos identificados. |
